Steps to enter DFU mode on your iPhone.
Step 1: Connect your iPhone to your PC or Mac and make sure that iTunes is running.
Step 2: Turn off the iPhone by holding the Power button and slide to power off
Step 3: Hold the power button for 3 seconds
Step 4: Next, you need to hold the Home and Power (sleep/wake) Buttons for about 10 seconds
Step 5: Then, release the Power button but keep pressing the home button for another 15 seconds
This will put your iPhone into DFU mode. When you connect the device to iTunes, a popup will tell you that iTunes has detected a device in DFU mode.

Part 2: How to Enter DFU Mode without Home Button or Power Button?
If for some reason you cannot use your home button or power button, you can still try to put iPhone in DFU Mode. The process is slightly more involving than the one above but it can be done.
How to put iPhone in DFU mode
Step 1: On your Desktop, create a folder that you will name Pwnage. In this recently created folder place the latest iOS firmware and the latest version of RedSn0w. You can download both online. Extract the RedSn0w zip file in this folder.
Step 2: Launch the extracted RedSn0w folder that was extracted earlier. You can do this very easily by right clicking on the .exe and selecting "Run as Administrator" from the contextual menu.
Step 3: Once the folder has been successfully opened, click on Extras
Step 4: From the Extra's menu in the resultant window, choose "Even More"
Step 5: From the Even More Menu in the resultant window select "DFU IPSW"
Step 6: A dialogue box will appear asking you to select an IPSW that you can currently restore to without any hacks. Click OK to continue
Step 7: Choose the ispw firmware file you downloaded in step 1 above and click Open
Step 8: Wait for DFU mode IPSW to be created
Step 9: A dialogue box confirming the successful creation of DFU mode IPSW will appear
Step 10: Next, Launch iTunes and connect your device to your computer. Select the device in the list on the left. If you have not performed a backup recently, this would be a good time to create on. Ensure you are on the Summary and then hold down the Shift Key and Click "Restore"
Step 11: In the next window, select "Enter-DFU ipsw from the folder we created in step One on your desktop and click "Open"
Step 12: This will put your iPhone In DFU mode. The screen will remain black and you may be able to jailbreak if you want depending on the firmware you chose.
